It would largely depend on what you're familiar with.
I would say RHEL 5.1 (already available) or Centos 5.1 (when it's out),
which comes with vendor-supported Xen 3.1.

(k)qemu works similarly to VMWare or VirtualBox.
Xen is entirely different.
